structural genomics

A

concerned with the organization and sequence information in a genome

how do you make a genomic library?

A
  1. different DNA molecules are cut in different places providing a set of overlapping fragments
  2. each fragment is joined to a cloning vector, transferred to bacterial cells
  3. this produces a set of clones containing overlapping genomic fragments (some contain gene of interest)
  4. bacteria can be spotted and screened with a probe to identify the candidate clones (like a southern)

linkage analysis

A

-looking at 2 different features of the genome that occur together because of their proximity to each other
(recombination frequency)

recombination frequency

A

equals recombinant gametes over total gametes
= recombinant progeny/total progeny
-measured in centimorgans (cM)

positional cloning

A

allows us to connect a gene of interest with a previously mapped linked gene (or marker)
(“chromosome walking”)

A

to detect the presence/absence of specific DNA sequences
