1
Q

What does each lymphocyte express

A

single antigen receptor specificity

2
Q

What do naive lymphocytes produce to help respond to many numbers of antigens

A

identical clones

3
Q

What are three stages of B-cell development

A

Maturation (Ag independent) (primary lymph)
Activation (Ag dependent) (second lymph)
Differentiation (Ag Dependent) (second lymph/tissues)

4
Q

What is necissary for maturation of B-cell

A

IL-7 contact with stromal cells

5
Q

What is one key component of lag phase

A

Colonial selection

6
Q

What is allelic exclusion

A

Blocked VDJ arrangement of other heavy chain allele

7
Q

What are some mechanisms of generation of specificity diversity

A
Genetic diversity
Combinatorial VDJ joining
Sloppiness
P and N- region nucleotide addition
Somatic hypermutation
7
Q

What is combinatorial diversity

A

The combination of one V region with different D and J regions, and the combination of different heavy chains with light

7
Q

Where does generation of specificity diversity occur

A

primary lymphoid organs

8
Q

What is somatic hypermutation and where does it occur, key characteristics

A

Occurs in germinal center while B-cell divides in response to antigen.
Cells are rapidly dividing increasing chances of mutation, if mutation is bad the cell will die. Affinity will increase and colonal expansion will occur.
Fine tuning of antigen binding site of b-cells
Doesn’t change what B-cell recognizes

9
Q

VDJ makes up what region of antibody

A

Variable region

10
Q

What region is DNA mutated at a high rate at during somatic hypermutation

A

Complementary determining regions

11
Q

How do you determine combinatorial association and where does it happen

A

(VxDxJ heavy chain) x (VxJ light chain) happens in primary lymp tissues

12
Q

Where does somatic hypermutation occur

A

Second lymph tissue after it’s seen antigens

13
Q

How does the B cell choose membrane bound vs secreted

A

Differential splicing

14
Q

If a B-cell expresses IgA, what does it secrete

A

IgA

15
Q

How is IgM to IgD class switched

A

RNA

differential splicing

16
Q

How is IgM to IgG, IgE, or IgA class switched

A

DNA differentiation

17
Q

What chemical determines the class switching that antibody needs to occur

A

Cytokines secreted by PAMP

18
Q

How many types of antigen does a B-cell recognize

A

1

19
Q

Does class switching change the specificity of the paratope

A

No

20
Q

What does class switching change

A

constant region

how the antibody will interact with everything
